    Investigation and optimization of mixed-mode chromatography for the analysis of pharmaceutical analytes dissolved in neat or high content of organic solventThesis project: 30 creditsStudent location: AstraZeneca, Gothenburg, SwedenStart: January 2022Supervisors: Ass. Prof. Morgan Stefansson morgan.stefansson@astrazeneca.com, Senior Scientist Anna Granfors Anna.granfors@astrazeneca.com BackgroundAt AstraZeneca a robot allowing automatic sample preparation has been developed and is also hyphenated to a chromatographic separation system for analysis. Methanol is used as dissolving medium creating a mismatch to the generally employed reversed phase systems. After initial tests, using a combination of fundamentally different retention mechanisms of the separation column, results showed great potential for its use. The separation column is packed with particles enabling hydrophobic as well as ion exchange interactions, usually called mixed-mode, and by a proper choice of chromatographic conditions the sample incompatibility can be overcome.Project planThe objective of this thesis plan is to investigate chromatographic parameters available, e.g.Mobile phase composition and gradient elution settings (ionic strength, composition, pH, slope etc.)Type of separation mediumSeparation selectivity and analyte detectability, including injection volumeRobustnessAnalysis time for high sample throughputAfter screening of these and other parameters, an optimization aiming for a general procedure and fitting the automatic sample preparation system would be a great step forward. Finally, an application of a drug product would prove the case.

For these dysfunctions effective pharmaceutical treatments are lacking, which causes  chronic pain, disabilities, and sudden death in patients. We aim to address this unmet medical need by uncovering core mechanisms of disease initiation and maintenance and leveraging this information for establishing novel therapeutic strategies for VAs. Our pre-clinical studies, testing identified treatment strategies are relevant for VAs and for understanding aberrant vascular function in vascular-related diseases. We are working with cutting-edge technologies including next-generation sequencing, CRISPR-Cas genome editing, generation of induced pluripotent stem cells (iPSCs) and animal models and in vivo phage display to identify endothelial targets.This position is within the Transgenic Mouse Model group which is part of the Discovery Biology (DB) department. Within the Transgenic unit, we aim  to be at the scientific forefront of generating and phenotypically characterizing transgenic mouse models as well as  performing world leading science and innovation.
